This antibody is predicted to react with bovine, canine, chicken, human, mouse and non-human primate based on 100% sequence homology. It does not cross-react with other purified substrates of cdk5 (amphiphysin and synapsin). This antibody is specific for the ~95 kDa Dynamin protein phosphorylated at Ser774in Western blots of rat brain lysates. This antibody labels the purified protein phosphorylated in vitro by cdk5 but not by PKC. Immunolabeling is blocked by the phosphopeptide used as the antigen but not by the corresponding dephosphopeptide. Store at -20ºC short term, 80ºC long term. Dynamin is a GTPase required for endocytosis of clathrin coated vesicles from the membrane. Dynamin's involvement in endocytosis was first discovered during the characterization of the temperature sensitive parylytic shibire mutation from Drosophila melanogaster. Three closely related isoforms have been identified in mammals. Dynamin1 is expressed exclusively in neurons, dynamin2 is ubiquitously expressed and dynamin3 is expressed primarily in the testes. Each of these isoforms of dynamin also has several splice variants.
